Telomeric Repeat Amplification Protocol (TRAP)

Recent advances in the TRAP assay:

New Cx reverse primers by Kim & Wu (Nucleic Acids Research 25:3595-2597, 1997) reduce primer dimer problems:

ACT primer: GCGCGG[CTAAACC]3
ACX primer: GCGCGG[CTTACC]3CTAACC

Redesigned primers by Miura et al.(Biochem. Biophys. Res. Comm. 246:13-19, 1998) may be more sensitive and semi-quantitative:

(first 25 cycles) forward primer: ATTGCCAATCCGTCGAGCAGAGTT
(last 2 cycles) forward primer: 32P-GCCAATCCGTCGAGCAGAGTTAGGG
(all cycles) reverse primer: CTCCC[TAACCC]3
